If you keep reaching a wall you cannot climb or a marked floor you cannot break, another weapon upgrade will not solve the problem. Dead Cells uses permanent movement runes to open routes, and several of those runes sit away from the familiar path through the Ramparts and the Concierge.

The useful question at the start of a run is: which rune am I missing, and which exit gets me to it? This guide follows one base-game route with no active Boss Stem Cells, usually called 0BC. It needs no paid DLC. Other routes can exist with DLC or higher difficulty; they are not required for this plan.

Choose the next target from the runes you already own

Your current progress Next target Route from Prisoners' Quarters
No movement runes Vine Rune Promenade of the Condemned
Vine, but no Teleportation Teleportation Rune Toxic Sewers
Vine and Teleportation, but no Ram Ram Rune Promenade of the Condemned → Ossuary
Vine, Teleportation and Ram, but no Spider Spider Rune Toxic Sewers → Ancient Sewers → Insufferable Crypt → Slumbering Sanctuary

In the final biome, defeat the rune’s elite guardian and pick up the reward; reaching the biome alone does not unlock it. Continue your current run and choose the next target when you next start from the prison.

These are separate target runs, each starting in Prisoners' Quarters. Do not read the table as one continuous journey from the Vine Rune straight to the other three. After a successful pickup, you can continue your current run; choose the next target when you next start from the prison.

Runes unlock immediately when collected and remain available in later runs. You do not need to carry them to the Collector, pay cells for them, or finish the game to keep them. The main menu's save-slot information also shows which runes that save has collected.

First target: Vine in the Promenade

Leave Prisoners' Quarters through the Promenade of the Condemned. In the Promenade, find the special encounter with an Elite Undead Archer, defeat it and pick up the Vine Rune.

Vine lets you grow climbable vines from the appropriate plants. Its important next step for this plan is the Toxic Sewers entrance back in Prisoners' Quarters on a later run.

Treat the pickup as the objective, not merely winning an elite fight. Check the reward before moving on. A rune guardian is a specific encounter; defeating another strong enemy elsewhere does not substitute for collecting its rune.

Once Vine is yours, continue with the equipment and health you have. There is no need to throw away the rest of a promising run. The permanent unlock has already made the attempt productive, even if the later boss still defeats you.

What changes on your next visit to the prison

At the beginning of the next run, stop before automatically choosing the Promenade again. Your destination is now Toxic Sewers. The new ability is valuable because it changes your route choice, not because the familiar exit has disappeared.

If you return to the Promenade by habit, that can still be a normal run, but it is not the Teleportation target route. Decide whether you are practising that route or pursuing the next unlock before selecting the exit.

Second target: Teleportation in Toxic Sewers

Use Vine to reach the Toxic Sewers entrance from Prisoners' Quarters. In the sewers, locate the special Elite Slasher encounter and collect the Teleportation Rune after defeating its guardian.

The new rune activates teleportation monoliths—the statues that previously did not provide a usable route. For this unlock sequence, their next important destination is the Ossuary entrance in the Promenade.

Do not assume that acquiring a new ability means its next target is farther ahead on your current path. Toxic Sewers has several exits, but the route to your first Ram Rune in this plan begins with a later visit to the Promenade.

Finish exploring what you need for the current run, then continue if you want to. On your next start, choose the Promenade rather than returning to Toxic Sewers just because that was where the last unlock happened.

A floor you cannot break is not a failed Teleportation unlock

Teleportation and Ram solve different obstacles. If a marked floor still blocks you after collecting Teleportation, that is expected. The new ability gets you to the biome that contains the floor-breaking rune; it does not itself break the floor.

This is the first useful dependency to remember: Teleportation opens the route to Ram. Repeatedly attacking a floor or changing weapons will not replace that missing step.

Third target: Ram in the Ossuary

Start with Prisoners' Quarters → Promenade of the Condemned. This time, do not take the familiar Ramparts exit automatically. Look for the route reached through a teleportation statue and check that its destination is the Ossuary.

The level layout is generated, so use the obstacle and destination name rather than expecting the doorway at a fixed screen coordinate.

Inside the Ossuary, find the special Elite Slasher encounter and pick up the Ram Rune. If you find the exit to Black Bridge first, keep searching for your target before leaving. Reaching the next boss without the rune does not complete this stage of the plan.

Ram lets your downward slam break the appropriate rune-marked floors. Use your platform's movement and jump controls for that action; an ordinary weapon attack is not the floor-breaking move.

You can now take the Ancient Sewers route from Toxic Sewers on a later run. Ram does not make the Ossuary itself lead backward into Toxic Sewers. Record the next starting choice before the current run's later encounters distract you.

Example: you own Teleportation but keep missing Ram

Suppose you have visited the Promenade several times, taken the Ramparts each time, and eventually fought the Concierge. Nothing is wrong with Teleportation. You have been following a route that skips the target biome.

On the next attempt, make the Promenade exit search the priority. Find the statue route, read “Ossuary,” and only then commit to leaving. Once there, make the guardian pickup—not the first available exit—the completion check.

That gives you two concrete checkpoints to correct: entering the right biome and collecting the right reward. It is more useful than repeating the entire familiar run and hoping the rune appears somewhere new.

Fourth target: Spider through the sewer boss route

With Ram collected, begin the longer route:

  1. Prisoners' Quarters → Toxic Sewers, using Vine for access.
  2. Toxic Sewers → Ancient Sewers, using Ram at the required floor passage.
  3. Ancient Sewers → Insufferable Crypt.
  4. Defeat Conjunctivius in the crypt.
  5. Choose Slumbering Sanctuary and find its Elite Caster guardian.
  6. Defeat the guardian and collect the Spider Rune.

Toxic Sewers and Ancient Sewers are different biomes. The former contains the Teleportation target; the latter is the next stage leading to Conjunctivius. Read the exit name rather than treating any sewer route as equivalent.

Likewise, Insufferable Crypt is the boss stop on this path. Seeing Black Bridge instead means you are on the Concierge branch, not the first-Spider route described here.

Why the Sanctuary door after the Concierge seems circular

The entrance to Slumbering Sanctuary from Black Bridge already requires Spider. That door is useful after you own the rune; it is not the entrance you use to obtain your first one on this base-game 0BC route.

The solution is to approach from Insufferable Crypt. After Conjunctivius, choose Slumbering Sanctuary rather than trying to force a passage that asks for the ability you are still seeking. You do not need to buy DLC to resolve this apparent loop.

This is the main reason to plan the route at Prisoners' Quarters. By the time you are standing beyond the Concierge, the early branching choice has already sent that run somewhere else.

In the Sanctuary, distinguish the rune hunt from opening the exits

Slumbering Sanctuary initially has inactive statues and orange doors blocking parts of the biome. Activating the Ancient Temple Switch awakens the area: the doors open and statues become active enemies as you approach.

Do not read an orange barrier as proof that you need another movement rune. The switch changes the biome's state. Your permanent unlock objective is the Elite Caster's Spider Rune; the switch is part of navigating the Sanctuary and continuing onward.

After awakening it, reassess corridors you previously crossed rather than assuming the return path remains quiet. Move through the changed area deliberately, especially if the guardian fight has left you low on health.

Once collected, Spider lets you cling to walls. That is the point at which the previously inaccessible climbing routes become part of your future choices. You do not have to complete the remainder of the run for the rune to count.

If the guardian no longer appears

If the relevant guardian encounter no longer appears, check the save's collected runes before searching the whole biome again. The special rune rooms stop generating once their rune has been obtained. A missing repeat guardian can be evidence of completed progress, not a broken save.

Keep each attempt focused without discarding a good run

Before leaving Prisoners' Quarters, name one target and its critical branch. For Ram, that is the Ossuary exit in the Promenade. For Spider, it is Ancient Sewers from Toxic Sewers and then the crypt's Sanctuary exit.

Before leaving the target biome, confirm the pickup. Afterward, continue playing rather than treating the rest of the attempt as wasted time. You can learn the next encounters while keeping the permanent progress already earned.

On a later run, choose from your updated list of owned runes—not from the route that happened to work last time. That turns exploration into a clear sequence of unlocks while leaving you free to enjoy the run between them.
